■ 研究概要

◆研究課題平滑筋収縮調節メカニズムおよび平滑筋機能疾患の病態解明 ◆研究概要平滑筋組織は，末梢臓器および脈管系の管壁を構成しており，胃腸管および泌尿・生殖器の運動，気道抵抗の調節，血圧の調節といった様々な生理機能を担っている。平滑筋の収縮活性は，神経伝達物質やホルモンによって緻密に制御されている。これら内因性情報伝達物質は，平滑筋細胞に発現する受容体と結合して，それぞれの受容体に固有の情報伝達機構を作動させる。これにより，細胞に発現するCa2+透過性イオンチャネルの活性が変化する結果，細胞内のCa2+濃度が変化し，最終的に筋の収縮活性が変化する。また，平滑筋組織の伸展が機械刺激感受性チャネルを介して筋の収縮活性を変化させることも知られている。平滑筋組織の形態および機能的変化は，構成する臓器の機能異常につながる。平滑筋機能障害が原因となる疾患の例として，過敏性腸症候群や過活動膀胱が挙げられる。本課題では，未解明な点が未だ多く残されている①平滑筋収縮調節メカニズムおよび②過敏性腸症候群や過活動膀胱をはじめとする平滑筋機能疾患の病態について研究を行う。上記の点が明らかになれば，関連する疾患に対する新たな治療薬の開発に必要な基盤情報を提供することができる。本年度も，小腸平滑筋の収縮調節におけるPiezo1チャネルの役割およびその収縮調節メカニズムについて研究を行う。また，マウス膀胱平滑筋のムスカリン作動性収縮におけるCa2+動員機構についても引き続き研究を行う。今年度はTRPCチャネルに加え，TRPMチャネル，Ano1チャネル，Oraiチャネルの関与についても検討する。さらに，過敏性腸症候群の病態解明に関する研究にも着手する予定である。
